Det framställs av 5,10-metylentetrahydrofolat av antingen ett NAD+-beroende metylentetrahydrofolatdehydrogenas eller ett NADP+-beroende dehydrogenas . Det kan också produceras som en mellanprodukt i histidinkatabolism , av formiminotransferascyklodeaminas , från 5-formiminotetrahydrofolat .

5,10-CH=THF är ett substrat för metenyltetrahydrofolatcyklohydrolas , som omvandlar det till 10-formyltetrahydrofolat .
